$CAPR Adcomm shows the rigor of FDA reviews. Clear communication with FDA throughout the process is critical to avoid confusion. FDA review is far more rigorous than peer reviewed journals, including the Lancet. Pre-specified analyses are sacred & largely ignored in peer review.
@AppleHelix In regulatory parlance, if FDA says "no discussion held, no agreements reached" that means it might as well have never existed. I'm not saying that's right, just that this is how FDA works.

@AppleHelix The issue is not with the data or the imputation method. The issue is that FDA needs to align with the company prior to an SAP to be valid, especially if changes made can impact endpoints. What $CAPR did unfortunately gives FDA a clear "out" to not approve.
@AppleHelix You are right: there is no requirement. But if there is no alignment in writing, it gives FDA the room it needs to reject. This is part of the art of working with FDA. Unfortunately, even strong science isn't black and white.
$CAPR can change their SAP all they want prior to unblinding, but the changes need to be approved by FDA to have any validity. Without FDA's approval, the SAP change is not valid.
